
Instrumentation
1. 2D off-line nano-LC workstation with micro fraction collector (Agilent Technologies).
This instrument combines strong cation exchange (SCX) and reversed phase (RP) chromatography. SCX chromatography is performed using a capillary pump by applying a linear salt gradient with subsequent fraction collection using the novel Agilent micro fraction collector. The following RP chromatography for every fraction collected is performed using a nano-pump by applying a gradient of increasing organic solvent concentration.
2. ABI 4000 Q TRAP LC/MS/MS System.
This instrument combines triple quadrupole scan modes with ion trap scans in a single LC/MS/MS run and has many proteomic applications. We are using the instrument for triple quadrupole multiple reaction monitoring (MRM) at the highest level of sensitivity and extended dynamic range.
3. ABI 4700 MALDI-TOF/TOF System with Dionex Probot spotter.
This instrument combines a MALDI source with a TOF/TOF tandem mass analyzer to acquire both peptide mass fingerprint data and MS/MS spectra of peptide ions via high energy collisionally-induced dissociation. Unique automation capabilities of the ABI 4700 provide for high-throughput analyses and enable off-line analysis of LC-fractionated peptide mixtures.
4. Various protein and peptide separation techniques include:
preparative and 2D-gel electrophoresis, gel elution, IEF fractionation in solution, AKTA-FPLC, and density gradient fractionation with Trumpet Tip.
